STANDARD OPERATING PROCEDURE Title: Preparation of Nutrient Agar

1. Purpose

a) It is established to provide a standard procedure for preparation of Nutrient agar.

2. Scope

a) It is applicable for Microbiological Testing.

3. Responsibilities

a) QC Manager

b) Microbiologist

4. Procedure

a) Take a sterilized 500ml screw capped conical flask and take 500ml of D/water in it.

b) Warm it about boiling.

c) Weigh 14 gm of media on butter paper and pour it into flask containing D/water.

d) Swirl it to dissolve while heating, until a clear solution is obtained.

e) Autoclave the media for 18 minutes at121^oC and 15lb pressure.

f) After autoclaving cool the media bottle to about 50^oC.

g) Take sterilized petri-dishes and pour the media in into them.
